Recombinase-based heat-inducible sensor

This part is a recombinase-based heat-inducible sensor. Under high-temperature conditions, RNA polymerase binds to the heat shock promoter (HSP) and begins transcription. The translated Bxb1 integrase protein then flips the DNA region flanked by recombinase sites attB and attP, irreversibly flipping a constitutive promoter to begin transcription of whatever downstream actuator is placed after the part.

Usage and Biology

The part serves as a binary memory device to record whether or not the organism has ever been exposed to temperatures above a certain threshold and can be coupled to the output of the user's choice. A potential application of the technology would be in a temperature sensor for food items that may be sensitive to exposure to high temperatures. An easy detected readout (GFP, RFP) could signal to the user or purveyor when a good has been exposed to high temperatures (perhaps sitting in a warm truck if refrigeration failed) and alert them to potential food safety concerns. A well-characterized heat-inducible promoter was used so that there should be a clear difference in expression from 4 C (refrigeration temperatures) to 37 C and above.
